Arbetsuppgifter Postdoktorn förväntas utföra oberoende/semioberoende forskning som utnyttjar insamlingen av mutanta C. elegans-alleler och/eller Pilon-gruppens expertis för att ytterligare främja vår förståelse av lipid/membranhomeostas. Subject area Single-cell genomics and functional imaging of kidney epithelial injury Subject area description The project aims to develop and apply novel experimental methods to study functional heterogeneity in proximal tubular epithelial cells (PTECs) during kidney injury. By combining direct measurements of albumin reabsorption with single-cell transcriptomics, the project seeks to identify molecular programs underlying epithelial dysfunction and to discover targetable pathways relevant to kidney disease. Duties The successful candidate will lead experimental work across multiple PTEC injury models, including toxic, metabolic, inflammatory, and hypoxia–reoxygenation stress. Key tasks include functional uptake assays using fluorescently labelled albumin, cell and tissue imaging, preparation of single-cell RNA sequencing libraries, data generation from primary human PTECs and acute kidney slices, and close collaboration with computational researchers for downstream analysis. Validation of candidate pathways in tissue slices will be an additional component of the work. • Optimization of functional cell-based assays to study proximal tubular epithelial cell (PTEC) physiology and injury responses • Performing experiments in cell culture systems and acute kidney tissue slices • Measurement of albumin uptake and other functional readouts using fluorescence-based and imaging approaches • Preparation of single-cell RNA sequencing libraries using established platforms (e.g. SmartSeq2, 10X Genomics, Parse) • Generation and quality control of experimental data from functional assays, imaging, and sequencing experiments • Collaboration with computational researchers for downstream data analysis and interpretation • Design and execution of validation experiments using molecular biology and imaging-based methods • Documentation and optimization of experimental protocols • Presentation of results in group meetings, scientific conferences, and manuscript preparation • Mentoring of junior researchers and students Eligibility The eligibility criteria for employing teaching staff are set out in Chapter 4 of the Higher Education Ordinance and in the Appointment Procedure for Teaching Posts at the University of Gothenburg. To be eligible for appointment as a postdoc, the applicant is required to have a doctoral degree, a doctoral degree in art or a foreign degree that is deemed to be equivalent to a doctoral degree. The candidate will contribute to the improve Slice-seq, a new method to study human pancreatic tissue slices using high-resolution calcium imaging in combination with spatial transcriptomics. The researcher will develop methods to improve the viability of pancreatic slices cultures, develop approaches for their cryopreservation, and distribution. The researcher will also run single-cell genomic and spatial transcriptomic assays on pancreatic tissue. The project will require significant interdisciplinary work in the areas of physiology, molecular biology and islet biology. The research group is embedded in the Wallenberg Center for Molecular and Translational Medicine (WCMTM) and SciLifeLab in the University of Gothenburg. and is tightly connected to a broad network of international collaborators. Subject area Bioinformatics and Cancer Genetics Subject area description Somatic mutation accumulation and clonal evolution are central processes in cancer development. Through large-scale whole-genome sequencing and other high-throughput approaches, these processes can be mapped in detail, providing insights into tumor initiation, progression, and treatment response. This position focuses on the application and development of bioinformatics methods for the analysis of genomic cancer data, including large public international datasets and locally generated patient-derived sequencing data. The project may involve, for example, studies of somatic mutational processes, clonal dynamics, tumor evolutionary history, or integrative analyses of genomic and clinical data. Analyses of non-cancer-related sequencing data may also be considered. The work will be carried out in a research environment with strong expertise in cancer genomics, computational biology, and translational research. The project will explore the role of goblet cell-intrinsic mucosal defence systems in determining susceptibility to CRC. The candidate will join a newly established Wallenberg Centre for Molecular and Translational Medicine (WCMTM) at the University of Gothenburg (www.wcmtm.gu.se). As part of this venture, Dr. George Birchenough has established his research group (www.birchenoughlab.org) at the Department of Medical Biochemistry & Cell Biology at the University of Gothenburg. The group works towards a better understanding of how mucosal defence systems control our interactions with the environment, how those systems fail in disease and how they might be prophylactically or therapeutically targeted. The group is part of a large network of researchers at Gothenburg University, most importantly the Mucin Biology Groups cluster (www.medkem.gu.se/mucinbiology) that currently includes over 30 researchers working at the forefront of mucosal biology. Subject area Translational Mucosal Biology Subject area description Our mucosal surfaces are the interface between the environment and our tissues. Their physiological functions are critical, but necessitate constant exposure of living cells to a vast collection of microbial lifeforms that includes many opportunistic and obligate pathogens. As a result, our mucosa has evolved multiple defensive systems that allow them to endure in this environment, and failure of these systems is often linked to development of acute and chronic disease. Consequently, understanding the development and regulation of these systems is key to understanding the pathoaetiology of mucosal disease and how they can be prevented or treated. Colorectal cancer (CRC) is one of the most frequent and lethal types of cancer, and is especially prevalent in nations with a high human development index, indicating that factors associated with a modern, Westernized lifestyle are linked to CRC development. Most cases of CRC result from spontaneous mutations rather than heritable oncogenic mutations, and dysregulated interactions between the intestinal microbiota and immune system are implicated in tumorigenesis. Goblet cell-intrinsic defence systems, such as the mucus layer and sentinel goblet cells normally control host-microbiota interactions, but we have recently found that these systems are highly susceptible to environmental alterations caused by exposure to a Western-style diet. Thus, disruption of goblet cell-dependent functions may have a causal relationship with CRC development. By combining data from pre-clinical mouse models with analysis of samples from healthy and CRC patients, we hope to develop a better understanding of how goblet cell functions contribute to CRC susceptibility and develop new approaches to prevent CRC development. Duties The successful candidate will be expected to work extensively with pre-clinical mouse models and human tissues acquired during routine surgery. The candidate will be encouraged and expected to conduct their experimental research in an independent manner and to make significant contributions to project planning. Project description: Ulcerative colitis is a chronic inflammatory bowel disease characterised by periods of remission and relapse. Although modern treatment often leads to clinical, endoscopic and histological healing, many patients relapse. Why an apparently healed intestinal mucosa remains biologically vulnerable is a central unanswered question. This PhD project aims to identify the molecular mechanisms that determine whether remission remains stable or the disease relapses. The project is based on the hypothesis that the intestinal microenvironment, shaped by interactions between the microbiota, microbial metabolites and host regulatory signals, is not fully restored during remission, which may increase susceptibility to renewed inflammatory stimuli. The PhD student will be part of BioLabX, Sweden's national research school in Biomedical Laboratory Science. The project is particularly relevant for applicants with a background in Biomedical Laboratory Science. The PhD student will combine patient-oriented research with experimental models to investigate how the intestinal microenvironment influences immune function during remission. The work includes handling and analysis of human patient samples, functional cell models using human macrophages, advanced cell culture, flow cytometry and molecular biology, as well as metabolomics and lipidomics. Bioinformatic and statistical methods will be used to integrate molecular data with clinical follow-up and identify biomarkers that may predict relapse. The research environment is translational and interdisciplinary, with close collaboration between preclinical researchers, clinical gastroenterologists and national research networks. Through BioLabX, the PhD student will have access to a national network, specialised doctoral courses and pedagogical training. Project description: Synapses relay flow of information between neurons and constitute the minimal processing units of our brains. Given their importance for brain computations, synaptic imbalances are thought to underlie a majority of neuropsychiatric, psychiatric, and neurodevelopmental diseases. A large fraction of the brain’s synapses undergoes controlled removal during development, but formation and balanced removal of synapses continue throughout life, presumably to support learning and memory. An excessive loss of synapses during late adolescence is thought to underlie schizophrenia. However, while synaptic transmission is well-characterized at the molecular level, much less is known about the cell biological processes that govern their structural assembly and disassembly. The Sterky lab aims to identify key molecular steps during synapse assembly, stabilization and disassembly (see https://sterkylab.se). For this, we focus on synaptic cell adhesion molecules, proteins on the neuron’s surface that physically interact with the contacting neuron, the proteins they in turn interact with and means by which they may be regulated. Several of these proteins are genetically linked to human diseases such as schizophrenia, and the long-term goal of this research is to develop means to normalize synaptic imbalances that underlie human disease. This project will address the synaptic role of a specific protein family by experimentally analyzing both genetically modified human neurons and mice. An array of experimental techniques will be used, including advanced cell culture, CRISPR/Cas9 gene editing, protein and transcript analysis, confocal microscopy and assessment of synaptic functions. Previous experience from work with laboratory animals and/or electrophysiological recordings would be considered an asset.
